Synergistic Interactions of Blood-Borne Immune Cells, Fibroblasts and Extracellular Matrix Drive Repair in an In Vitro Peri-Implant Wound Healing Model
The authors showed that alkali-treatment, compared to native titanium surfaces, increased blood clot thickness, including platelet adhesion. Blood clots with entrapped blood cells in synergistic interactions with fibroblasts, but not fibroblasts alone, upregulated the secretion of major factors associated with fast healing. Researchers provide evidence that overexpression of Cyclin D1 induces reprogramming of epidermal cells into stem cell-like cells. [Cell Cycle] Abstract

Estrogen Promotes Healing in a Bacterial LPS Model of Delayed Cutaneous Wound Repair
Investigators asked whether estrogen would effectively promote healing in an LPS bacterial infection model. Co-treatment with 17β-estradiol was able to promote re-epithelialisation, dampen inflammation and induce collagen deposition in our LPS-delayed healing model. [Lab Invest] Abstract

Human Amniotic Epithelial Stem Cells Promote Wound Healing by Facilitating Migration and Proliferation of Keratinocytes via ERK, JNK and AKT Signaling Pathways
Researchers found that human amniotic epithelial stem cells (HAESC) accelerate the migration of keratinocytes and induce a remarkable increase in the activity of phospho-ERK, phospho-JNK, and phospho-AKT, the blockade of which by their specific inhibitors significantly inhibited migration induced by HAESC-conditioned medium. Hexosamine Biosynthesis in Keratinocytes – Roles of GFAT and GNPDA Enzymes in the Maintenance of UDP-GlcNAc Content and Hyaluronan Synthesis
Depletion of glutamine-fructose-6-phosphate aminotransferases (GFAT) reduced the cellular pool of UDP-N-Acetylglucosamine (UDP-GlcNAc) and hyaluronan synthesis, while simultaneous blocking of both glucosamine-6-phosphate deaminases (GNPDA) exerted opposite effects, indicating that in standard culture conditions keratinocyte GNPDAs mainly catalyzed the reaction from glucosamine-6-phosphate back to fructose-6-phosphate. Changes in Dermal Matrix in the Absence of Rac1 in Keratinocytes
The authors assessed the effects of Rac1 deletion in keratinocytes and of the heightened inflammatory status by induction of irritant contact dermatitis on the tissue localisation and arrangements of dermal collagen. [J Anat] Abstract


XPA Promotes Autophagy to Facilitate Cisplatin Resistance in Melanoma Cells through the Activation of PARP1
Scientists found that xeroderma pigmentosum (XPA) was expressed at higher levels in cisplatin-resistant melanoma cells than that in cisplatin-sensitive ones. The knockdown of XPA not only increased cellular apoptosis but also inhibited cisplatin-induced autophagy, which rendered the melanoma cells more sensitive to cisplatin. [J Invest Dermatol] Abstract

SOX9 Indirectly Regulates CEACAM1 Expression and Immune Resistance in Melanoma Cells
Investigators examined the role of transcription factor SOX9 in the regulation of CEACAM1 (carcinoembryonic antigen cell adhesion molecule 1) expression and immune resistance in melanoma cells. [Oncotarget] Full Article

Ubiquitin Ligase UBE3C Promotes Melanoma Progression by Increasing Epithelial-Mesenchymal Transition in Melanoma Cells
Researchers analyzed ubiquitin-protein ligase E3C (UBE3C) expression in samples of cancerous and normal skin tissue. Levels of UBE3C expression were much higher in primary and metastatic melanoma tissues than in normal skin, cutaneous squamous cell carcinoma or basal cell carcinoma. [Oncotarget] Full Article
