Galectin-7 Regulates Keratinocyte Proliferation and Differentiation through JNK-miR-203-p63 Signaling
Researchers showed that galectin-7 regulates keratinocyte differentiation and proliferation through microRNA (miR)-203 but not miR-146a. A knockdown of either galectin-7 or miR-203 in keratinocytes increased expression of p63, an essential transcription factor involved in skin development. [J Invest Dermatol] Abstract

Differential Features Between Chronic Skin Inflammatory Diseases Revealed in Skin-Humanized Psoriasis and Atopic Dermatitis Mouse Models
The authors described and characterized an animal model for atopic dermatitis using similar bioengineered-based approaches, by intradermal injection of human T helper type 2 lymphocytes in regenerated human skin after partial removal of stratum corneum. [J Invest Dermatol] Abstract

Hypermethylation of the Keap1 Gene Inactivates Its Function, Promotes Nrf2 Nuclear Accumulation, and Is Involved in Arsenite-Induced Human Keratinocyte Transformation
By establishing transformed human keratinocyte cells via chronic arsenite treatment, researchers observed a continuous reduction in reactive oxygen species levels and enhanced levels of nuclear factor E2-related factor 2 (Nrf2) and its target antioxidant enzymes in the later stage of arsenite-induced cell transformation. Hair Follicle and Interfollicular Epidermal Stem Cells Make Varying Contributions to Wound Regeneration
By comparing the fates of several keratinocyte lineages, scientists observed an initial decrease in both hair follicle- and interfollicular epidermis-derived cells within the transient acanthotic layers of the regenerating epithelium. [Cell Cycle] Abstract

The Proteasome Immunosubunits, PA28 and ER-Aminopeptidase 1 Protect Melanoma Cells from Efficient MART-126-35 Specific T-Cell Recognition
Researchers demonstrated that the IFN-γ-inducible proteasome subunit β2i/MECL-1, proteasome activator PA28 and ER-resident aminopeptidase ERAP1 impair MART-126-35 epitope generation. [Eur J Immunol] Abstract

The Wound Healing and Antibacterial Activity of Five Ethnomedical Calophyllum inophyllum Oils: An Alternative Therapeutic Strategy to Treat Infected Wounds
Using cell and bacteria cultures, investigators confirmed the pharmacological effects of Calophyllum inophyllum oil (CIO) as wound healing and antimicrobial agent. They showed that the concentration of CIO needed to exhibit therapeutic effects are lower than concentrations exhibiting cytotoxic effects in vitro. [PLoS One] Full Article

Pharmacological Activation of Estrogen Receptors-α and -β Differentially Modulates Keratinocyte Differentiation with Functional Impact on Wound Healing
Investigators revealed that the HaCaT keratinocytes expressed both estrogen receptor (ER)-α and ‑β, and thus are well-suited for studying the effects of ER agonists on epidermal regeneration. [Int J Mol Med]

GLUT10 Deficiency Leads to Oxidative Stress and Non-Canonical αvβ3 Integrin-Mediated TGFβ Signaling Associated with Extracellular Matrix Disarray in Arterial Tortuosity Syndrome Skin Fibroblasts
To discern the molecular mechanisms underlying the arterial tortuosity syndrome etiology, scientists performed gene expression profiling and biochemical studies on skin fibroblasts. Transcriptome analyses revealed the dysregulation of several genes involved in TGFβ signaling and extracellular matrix homeostasis as well as the perturbation of specific pathways that control both the cell energy balance and the oxidative stress response. Lysyl Hydroxylase 3 Localizes to Epidermal Basement Membrane and Is Reduced in Patients with Recessive Dystrophic Epidermolysis Bullosa

Particular S100 proteins differ in their distribution in epidermal layers, skin appendages, melanocytes and Langerhans cells. Taking into account that each epidermal component consists of specialized cells with well-defined functions, such differential distribution may be indicative of the function of a given S100 protein.
