UCLA Researchers’ Discovery Is Milestone in Understanding Treatment-Resistant Melanoma
Scientists studied drug resistance by analyzing samples of patients’ melanoma tumors both before they underwent therapy and when the disease recurred. The scientists also replicated the process outside of the human body — modeling drug resistance by growing melanoma cell lines from patients’ tumors and adapting them to drugs that block the MAP kinase pathway. Integrin-Linked Kinase Regulates the Niche of Quiescent Epidermal Stem Cells
Researchers showed that integrin-linked kinase is a key regulator of the bulge extracellular matrix microenvironment, thereby governing the activation and maintenance of hair follicle stem cells. [Nat Commun] Full Article

Type VI Collagen Regulates Dermal Matrix Assembly and Fibroblast Motility
Researchers examined the expression pattern of type VI collagen in normal and wounded skin and investigated its specific function in new matrix deposition by human dermal fibroblasts. [J Invest Dermatol] Abstract

Global Gene Expression Analysis in PKCα−/− Mouse Skin Reveals Structural Changes in the Dermis and Defective Wound Granulation Tissue
The authors present data linking protein kinase C alpha (PKCα) to the regulated expression of multiple extracellular matrix components including small leucine-rich proteoglycans. [J Invest Dermatol] Abstract

Histamine Released from Epidermal Keratinocytes Plays a Role in α-Melanocyte-Stimulating Hormone-Induced Itching in Mice
The authors investigated whether α-melanocyte-stimulating hormone (α-MSH) elicited itch-related responses in mice. They found that an intradermal injection of α-MSH induced hind-paw scratching, an itch-related response, in mice. [Am J Pathol] Abstract

The E3 Ligase Itch Knock-Out Mice Show Hyper-Proliferation and Wound Healing Alteration
Scientists investigated the contribution of Itch in skin development and wound healing. Itchy new born mice manifested hyperplastic epidermis, which is not present in adulthood. Itch-/- cultured keratinocytes showed over expression of proliferating markers, increased capability to proliferate, migrate and to repair a scratch injury in vitro. [FEBS J] Abstract

Maintenance of Sweat Glands by Stem Cells Located in the Acral Epithelium
Researchers identified the specific stem cells in the acral epithelium that participate in the long-term maintenance of sweat glands, ducts, and interadnexal epidermis and that facilitate the regeneration of these structures following injury. [Biochem Biophys Res Commun] Full Article


NF-kB2 Induces Senescence Bypass in Melanoma via a Direct Transcriptional Activation of EZH2
The authors showed that the non-canonical-NF-kB pathway accounts for most of the NF-kB activity in melanoma cells, in contrast to non-cancer cells. They identified the non-canonical-NF-kB pathway as a key regulator of EZH2 expression in melanoma. [Oncogene] Abstract

A Novel Fully-Humanized 3D Skin Equivalent to Model Early Melanoma Invasion
Scientists developed a robust and reproducible, fully-humanized 3D skin equivalent comprising a stratified, terminally differentiated epidermis and a dermal compartment consisting of fibroblast-generated extracellular matrix. [Mol Cancer Ther] Abstract

Development and Characterization of a Novel In Vitro Progression Model for UVB-Induced Skin Carcinogenesis
Investigators demonstrated that ultraviolet B (UVB)-transformed human epidermal keratinocyte cells gain enhanced proliferation rate, apoptosis-resistance, and colony- and sphere-forming abilities in a progressive manner. [Sci Rep] Full Article

Focal Adhesion-Independent Integrin αv Regulation of FAK and c-Myc Is Necessary for 3D Skin Formation and Tumor Invasion
Researchers performed a multiplexed shRNA screen to define roles for each subunit in human organotypic skin. They showed that αv heterodimers are essential for epidermal generation, with αv loss driving a keratinocyte G1-S cell cycle block. [J Cell Sci] Abstract

Absence of RIPK3 Predicts Necroptosis Resistance in Malignant Melanoma
Scientists investigated the impact of inhibitor of apoptosis (IAPs) proteins for cell death regulation in malignant melanoma. Suppression of IAPs strongly sensitized a panel of melanoma cells to death ligand-induced cell death, which was largely mediated by apoptosis, as it was completely rescued by addition of caspase inhibitors. Cancer Research UK and MedImmune Launch Ground-Breaking Biotherapeutics Research Center in Cambridge
A new laboratory that will focus on the discovery and development of novel biologic cancer treatments and diagnostics has been opened in Cambridge. UAMS Scientist Awarded $1.7 Million Grant for Melanoma Research
University of Arkansas for Medical Sciences (UAMS) researcher Vladimir Zharov, Ph.D., D.Sc., was awarded a $1.7 million grant by the National Cancer Institute for clinical testing of a new technology called Theranostics, which is an integration of early diagnosis and treatment of melanoma.
