Exome Sequencing Identifies Recurrent Mutations in NF1 and RASopathy Genes in Sun-Exposed Melanomas
Scientists report on whole-exome sequencing of 213 melanomas. Their analysis established NF1, encoding a negative regulator of RAS, as the third most frequently mutated gene in melanoma, after BRAF and NRAS. Inactivating NF1 mutations were present in 46% of melanomas expressing wild-type BRAF and RAS, occurred in older patients and showed a distinct pattern of co-mutation with other RASopathy genes, particularly RASA2. Rapidly Light-Activated Surgical Protein Glue Inspired by Mussel Adhesion and Insect Structural Crosslinking
Researchers report a new light-activated, mussel protein-based bioadhesive (LAMBA) inspired by mussel adhesion and insect dityrosine crosslinking chemistry. LAMBA exhibited substantially stronger bulk wet tissue adhesion than commercially available fibrin glue and good biocompatibility in both in vitro and in vivo studies. [Biomaterials] Full Article

cAMP Promotes Cell Migration through Cell Junctional Complex Dynamics and Actin Cytoskeleton Remodeling: Implications in Skin Wound Healing
8-Bromo cAMP-treated mESCs showed significant wound closure and improved neovascularization. Moreover, 8-Bromo cAMP stimulated mESCs migration into the wound bed. [Stem Cells Dev] Abstract

Expression Profiles of Neuropeptides, Neurotransmitters, and Their Receptors in Human Keratocytes In Vitro and In Situ
Researchers assessed the endogenous intracellular and secreted levels of the neuropeptides substance P and neurokinin A, and of the neurotransmitters acetylcholine, catecholamines (adrenaline, noradrenaline and dopamine), and glutamate, as well as the expression profiles of their receptors, in human primary keratocytes in vitro and in keratocytes of human corneal tissue sections in situ. CCN1, a Pro-Inflammatory Factor, Aggravates Psoriasis Skin Lesions by Promoting Keratinocyte Activation
The authors demonstrated that CCN1, an extracellular matrix protein that is also considered a pro-inflammatory factor, is highly expressed in the lesional skin of psoriasis patients, as well as in that of imiquimod- and IL-23-treated psoriasis-like mice. [J Invest Dermatol] Abstract

Invasion of Herpes Simplex Virus Type 1 into Murine Epidermis: An Ex Vivo Infection Study
Investigators report how tissue has to be prepared to be susceptible to herpes simplex virus type 1 infection. Most efficient infection of the epidermis was achieved by removing the dermis. [J Invest Dermatol] Abstract

PAX3 and FOXD3 Promote CXCR4 Expression in Melanoma
Researchers found melanoma cells expressed two CXCR4 isoforms, the common version and a variant that is normally restricted to cells during development or to mature blood cells. [J Biol Chem] Abstract | Full Article

Unique MicroRNAs Appear at Different Times during the Course of a Delayed-Type Hypersensitivity Reaction in Human Skin
Scientists generated global microRNA expression profiles (using next-generation sequencing) of diphencyprone reactions in skin of seven healthy volunteers at three, 14, and 120 days after challenge. [Exp Dermatol] Abstract

VS-5584, a Novel PI3K-mTOR Dual Inhibitor, Inhibits Melanoma Cell Growth In Vitro and In Vivo
The authors evaluated the potential anti-melanoma effect of VS-5584, a novel and highly potent Phosphatidylinositol 3-kinase (PI3K)-AKT-mammalian target of rapamycin (mTOR) dual inhibitor. They demonstrated that VS-5584 potently inhibited survival and proliferation of established and primary human melanoma cells, but was non-cytotoxic to non-cancerous human skin keratinocytes and B10BR murine melanocytes.
