Histone H3K27 Demethylase JMJD3 in Cooperation with NF-κB Regulates Keratinocyte Wound Healing
Scientists report on the biological function of JMJD3 in keratinocyte wound healing using in vitro cell and in vivo animal models. Inactivation of JMJD3 or NF-κB resulted in aberrant keratinocyte wound healing. [J Invest Dermatol] Abstract | Full Article

Membrane-Tethered Intracellular Domain of Amphiregulin Promotes Keratinocyte Proliferation
Investigators showed that growth arrest of amphiregulin (AREG)-silenced keratinocytes occurs in G2/M and is significantly restored by full-length AREG precursor and AREG-cytoplasmic domains but not by AREG-extracellular domains. [J Invest Dermatol] Abstract | Full Article

Flexible pH-Sensing Hydrogel Fibers for Epidermal Applications
Scientists present pH-responsive hydrogel fibers that can be used for long-term monitoring of epidermal wound condition. [Adv Healthc Mater] Abstract

The Role of Oncogenic Ras in Human Skin Tumorigenesis Depends on Clonogenic Potential of the Founding Keratinocytes
Researchers investigated whether the oncogenic insult outcome may depend on the nature of the founding keratinocyte. They demonstrated that Ras-V12-overexpression induces senescence in primary human keratinocyte cultures, but some cells escape senescence and proliferate indefinitely. [J Cell Sci] Abstract

Plasminogen Is a Critical Regulator of Cutaneous Wound Healing
The authors report that plasminogen-deficient mice have extensive fibrin and neutrophil depositions in a wounded area long after re-epithelialization, indicating inefficient debridement and chronic inflammation. [Thromb Haemost] Abstract

Spatial Distribution of Stem Cell-Like Keratinocytes in Dissected Compound Hair Follicles of the Dog
To gain insight into the spatial distribution of follicular keratinocytes with stem cell potential in canine compound follicles, researchers microdissected hair follicles in anagen and telogen from skin samples of freshly euthanized dogs. [PLoS One] Full Article

Notch4 Signaling Induces a Mesenchymal-Epithelial-Like Transition in Melanoma Cells to Suppress Malignant Behaviors
Investigators report that expression of the constitutively active intracellular domain of Notch4 in melanoma cells triggered a switch from a mesenchymal-like parental phenotype to an epithelial-like phenotype. [Cancer Res] Abstract | Full Article

VEGF-A Acts via Neuropilin-1 to Enhance Epidermal Cancer Stem Cell Survival and Formation of Aggressive and Highly Vascularized Tumors
Researchers identified a limited subpopulation of epidermal cancer stem cells, in squamous cell carcinoma, that form rapidly growing, invasive and highly vascularized tumors, as compared with non-stem cancer cells. [Oncogene] Abstract

Directed De-Differentiation Using Partial Reprogramming Induces Invasive Phenotype in Melanoma Cells
Using a murine model, the authors investigated the effects of partial reprogramming on the cellular plasticity of melanoma cells. They demonstrated that induced partial reprogramming results in a reversible phenotype switch in melanoma cells. [Stem Cells] Abstract

MED27 Promotes Melanoma Growth by Targeting AKT/MAPK and NF-κB/iNOS Signaling Pathways
Scientists identified MED27 as a potential melanoma target and explored its role and associated molecular mechanism involved in melanoma progression. [Cancer Lett] Abstract

REVIEWS
The Role of Neutrophilic Inflammation, Angiotropism, and Pericytic Mimicry in Melanoma Progression and Metastasis
The authors discuss the role of neutrophilic inflammation in angiotropism and pericytic mimicry in melanoma progression, metastasis, tumor cell plasticity, and tumor therapeutic resistance. [J Invest Dermatol]
