Integrin Adhesions Suppress Syncytium Formation in the Drosophila Larval Epidermis
Scientists found that targeted knockdown of the integrin focal adhesion complex components β-integrin, PINCH, and integrin-linked kinase caused formation of multinucleate epidermal cells within the Drosophila larval epidermis. [Curr Biol] Abstract | Graphical Abstract

DERMAL STEM CELLS & TISSUE REGENERATION

The Human Papillomavirus (HPV) E7 Protein Antagonises an Imiquimod-Induced Inflammatory Pathway in Primary Human Keratinocytes
Researchers established that undifferentiated keratinocytes do not express the putative receptor for imiquimod, TLR7, and as such are stimulated by imiquimod through a novel pathway. [Sci Rep] Full Article

Secreted Biofilm Factors Adversely Affect Cellular Wound Healing Responses In Vitro
Staphylococcus aureus and Pseudomonas aeruginosa, were evaluated for their capacity to affect keratinocyte proliferation and migration. [Sci Rep] Full Article

Inhibition of Wnt/β-Catenin Pathway Promotes Regenerative Repair of Cutaneous and Cartilage Injury
In the naturally stented model of ear punch injury, researchers found that Wnt/β-catenin pathway is activated most notably in the dermis of the wound bed early after injury and subsides to baseline levels by d10. [FASEB J] Abstract

STAT3 Regulated ATR via MicroRNA-383 to Control DNA Damage to Affect Apoptosis in A431 Cells
The authors report that signal transducer and activator of transcription 3 (STAT3) inhibited DNA damage induced by UV and STAT3 mediated upregulation of GADD45γ and MDC-1 and the phosphorylation of H2AX in UV induced DNA damage. [Cell Signal] Abstract

TAT-Mediated Acidic Fibroblast Growth Factor Delivery to the Dermis Improves Wound Healing of Deep Skin Tissue in Rat
Researchers found that that mice treated with transactivator of transcription (TAT) protein-acidic fibroblast growth factor (aFGF) had higher accumulation of aFGF in both dermis and subcutaneous tissues compared with mice treated with aFGF alone. [PLoS One] Full Article

Investigating the Potential of Shikonin as a Novel Hypertrophic Scar Treatment
Scientists indicate that Shikonin preferentially inhibited cell proliferation and induced apoptosis in fibroblasts without affecting keratinocyte function. [J Biomed Sci] Full Article

SKIN CANCERS & DISORDERS

Targeted Inhibition of Metastatic Melanoma through Interference with Pin1-FOXM1 Signaling
The authors showed that the pro-proliferative transcription factor FOXM1 was elevated and activated in malignant melanoma. FOXM1 activity correlated with expression of the enzyme Pin1, which they found to be indicative of a poor prognosis. [Oncogene] Abstract

Fibroblast-Derived Neuregulin-1 Promotes Compensatory ErbB3 Signaling in Mutant BRAF Melanoma
The authors demonstrated that neuregulin 1 was highly expressed by dermal fibroblasts and cancer associated fibroblasts isolated from mutant BRAF melanomas. [J Biol Chem] Abstract | Full Article

TWEAK/Fn14 Activation Induces Keratinocyte Proliferation under Psoriatic Inflammation
Researchers explored the role of Tumor necrosis factor -like weak inducer of apoptosis (TWEAK)/fibroblast growth factor-inducible 14 (Fn14) pathway in the growth of psoriatic keratinocytes that is characterized by suppressed apoptotic cell death. [Exp Dermatol] Abstract

REVIEWS
Epithelial Cell Death and Inflammation in Skin
The authors review recent studies in genetic mouse models providing evidence that keratinocyte death is a potent trigger of skin inflammation and discuss their potential relevance for human inflammatory skin diseases. [Curr Top Microbiol Immunol] Abstract
