Researchers at Penn Develop Scar-Like Culture Systems to Understand and Treat Fibrosis
Researchers developed a “scar in a dish” in order to uncover mechanisms that cells use to sense this fibrous tissue. The system will be helpful for understanding the roots of the scars’ stiffness, and ultimately lead to treatments that would help revert the tissue to its normal consistency. Transcription Factor CTIP2 Maintains Hair Follicle Stem Cell Pool and Contributes to Altered Expression of LHX2 and NFATC1
Keratinocytes of the bulge stem cells niche of COUP-TF-interacting protein 2 (Ctip2)ep-/- mice proliferated more and underwent reduced apoptosis than the corresponding cells of wild-type mice. However, premature activation of follicular stem cells in mice lacking CTIP2 leads to the exhaustion of this stem cell compartment in comparison to Ctip2L2/L2 mice, which retained quiescent follicle stem cells. 7-Hydroxydehydronuciferine Induces Human Melanoma Death via Triggering Autophagy and Apoptosis
Investigators identified 7-hydroxydehydronuciferine isolated from the leaves of Nelumbo nucifera Gaertn cv Rosa-plena to be a bio-active agent that antagonizes melanoma tumor growth in mice xenograft model in vivo. [Exp Dermatol] Abstract

Modulating the Expression of Survivin and Other Basal Epidermal Proteins Protects Human Skin from UVB Damage and Oxidative Stress
The authors investigated the implication of survivin in epidermal renewal and the relationships between survivin expression and UVB-induced DNA damage levels in cultured human keratinocytes and in skin biopsies. [J Cosmet Dermatol] Abstract


CDKN2B Loss Promotes Progression from Benign Melanocytic Nevus to Melanoma
The authors showed that CDKN2B is highly upregulated in benign melanocytic nevi, contributes to maintaining nevus melanocytes in a growth-arrested premalignant state, and is commonly lost in melanoma. [Cancer Discov] Abstract

AMPK/HuR-Driven IL-20 Post-Transcriptional Regulation in Psoriatic Skin
Researchers showed that IL-20 was primarily upregulated in psoriatic skin at the post-transcriptional level. The RNA-binding protein HuR relocalized to the cytoplasm of keratinocytes of psoriatic patients, suggesting that it stabilizes numerous transcripts, as observed in the human keratinocyte cell lines used to assess IL-20 mRNA. miR-137 Inhibits Proliferation of Melanoma Cells by Targeting PAK2
The authors found that overexpression of miR-137 inhibited the proliferation of melanoma cells, which could be phenocopied by knockdown of p21-activated kinase 2 (PAK2) using siRNAs. [Exp Dermatol] Abstract

MicroRNA-375 Suppresses Extracellular Matrix Degradation and Invadopodial Activity in Head and Neck Squamous Cell Carcinoma
Increased miR-375 expression in head and neck squamous cell carcinoma cells suppressed extracellular matrix degradation and reduced the number of mature invadopodia. Higher miR-375 expression did not reduce cellular levels of selected invadopodia-associated proteins, nor was tyrosine phosphorylation of cortactin altered.
